HIGH COPY NUMBER PLASMID-LIKE EXPRESSION SYSTEM FOR PROTEIN PRODUCTION IN PLANTS
This technology provides a plasmid-like system for the conditional production of high levels of proteins in plants. Thus, it brings to plants genetic tools used until now only in microbes. It can be broadly used, both in monocot and dicot plants. This technology has significant advantages compared to existing technologies for protein expression in plants. Compared to the standard cloning of transgenes in the chromosome, it provides reproducibly higher levels of proteins because the gene of interest is expressed from a self replicating high-copy number episome. Production levels of up to ca. 5% of total protein are routinely expected. Compared to the current protein expression systems based on viruses, this system is inherently safer because it does not spread from cell to cell. It also has a much broader host range. Furthermore, this plasmid-like expression system is heritable and thus does not require a costly manual infection of the plant for the production of the protein. Finally, this expression system has the advantage of being inducible leading to a conditional production of the protein only in target tissue when desired.
