Immunogenic Compositions for Induction of Anti-Tumor Immunity
Immunotherapy of tumors is hampered by the physiological tolerance of the immune system to tumor associated antigens, like the p53 molecule, which are expressed in normal cells too. The p53 protein is the product of a tumor suppressor gene that functions to arrest the growth of mutated or aberrant cells. The majority of natural human tumors bear mutated or otherwise inactivated p53, which accumulate in the cytoplasm. Because tumor cells and normal cells differ in both amount and compartment of p53 expression, the p53 molecule may serve as target for immunotherapy. The present invention shows that activation of the immune system against the p53 protein and subsequent eradication of experimental tumors could be achieved by immunization with anti-p53 antibody, or peptides derived from the variable region of such antibody.
