Prevention Of Metastasis With 5-aza-2'-deoxycytidine
The present invention relates to methods for screening cancer patients for susceptibility to a particular treatment protocol and to a method of treatment of those patients found positive in the screen. Loss or inactivation of tumor suppressor genes is among the genetic defects which participate in the multi-step process that causes cancer. Some of the tumor suppressor genes are part of the genetic machinery which controls apoptosis and therefore their deregulation may lead to tumor growth. One of the tumor suppressor genes which is shut off in various cancer cells is DAP-kinase where the lack of DAP-kinase expression is caused by hypermethylation of the gene. In animal models DAP-kinase expression can be restored by treatment of cells with 5-aza-2'-deoxycytidine, a drug which inhibits DNA methylation. This suppresses the ability of the cells to form lung metastases. This invention is relevant to diagnosis in identifying cancer patients whose tumor cells have a hypermethylated DAP-kinase gene and therefore are susceptible for treatment with demethylating agents.
